Toulouse created its clearest opportunities during the first half but ran into an inspired Berke Özer.
The LOSC goalkeeper produced key saves to keep the match scoreless before halftime, while the hosts also struck the post.
Toulouse additionally appealed for a possible handball inside the penalty area. The incident was reviewed, but the VAR did not award a penalty.
Ayase Ueda scores Lille's winner
The hosts suffered a setback when goalkeeper Guillaume Restes was unable to continue.
Mathys Niflore was forced to replace him and eventually conceded the only goal of the match.
In the 73rd minute, Ayase Ueda broke the deadlock to give Lille a 1-0 advantage.
Toulouse, however, had opportunities to respond.
Shortly after Ueda's goal, Thomas Jørgensen found himself alone in front of goal but sent his effort over the bar.
He had another opportunity against Özer late in the match but once again failed to convert.
The Lille goalkeeper preserved his clean sheet and Davide Ancelotti's side secured all three points.
