Bublik blew the Australian out of the tournemtn, winning 6-7 (5-7) 6-4 7-5 in two hours and 22 minutes to reach his first Masters 1000 semi-final.
The 28-year-old from Kazakhstan struck 44 winners and did not drop his serve, improving to 2-2 in the pair's head-to-head series.
Bublik can still qualify for next month's Nitto ATP Finals but must win the title in Paris to do so.
Luckily for De Minaur, he had already booked his spot at the year-end event in Turin prior to this defeat, but will need to rebuild his confidence quickly if he is to have any success.
