American trio McGirt, Compton and Hoge carded eight-under-par 62s at Sedgefield Country Club in Greensboro, North Carolina on Thursday.
There were birdies aplenty in the final PGA Tour tournament before next week's opening FedExCup play-off event, with McGirt, Compton and Hoge tallying 22 between themselves.
The round of the day, however, belonged to Compton, who teed off on the back nine.
Compton - 114th in the FedExCup standings and already assured of a spot at The Barclays - birdied seven holes on the front nine, which included a span of four in a row to finish on a high.
It was the 35-year-old's lowest round of the season.
Jim Herman, Derek Ernst and Morgan Hoffman are a shot off the pace and tied for fourth.
FedExCup play-offs hopeful Tiger Woods enjoyed a welcome return to form to be just two shots adrift of the leading trio heading into the second round.
Playing the Wyndham Championship for the first time in his career, Woods posted his lowest opening round since 2012 via a six-under 64 on Thursday.
Woods - 187th in the standings with only the top 125 players advancing to The Barclays - needs to either win or finish outright second to have any chance of prolonging his season.
Martin Kaymer, Carl Pettersson and Davis Love III also registered 64s.
Tom Gillis, Jonas Blixt, Hideki Matsuyama, Bill Haas, Chad Campbell, Cameron Percy, Oscar Fraustro, George Coetzee and Sam Saunders are a shot further back.
Meanwhile, defending champion Camilo Villegas holed a four-under-par 66.
