The Dane led Humbert 6-4 2-2 when he felt a sharp pain in the limb and was forced to retire after one hour, 23 minutes.
Rune was in tears as he received treatment, and hopped off with the assistance of the medical team to sympathetic cheers from the crowd.
Rune won Stockholm in 2022 and looked good for a sixth straight win over Humbert, but it is the Frenchman who will now meet Casper Ruud in the final.
“It was not the way I wanted to win,” Humbert admitted after the match.
“I’m super disappointed for Holger, I hope he is going to be OK. I wish him a speedy recovery. It was a great match. He was playing a little bit better than me, but I’m really sorry for Holger.”
In the second semi-final, Ruud moved past Canadian Denis Shapovalov 6-3 6-4 to advance to his third final of the season after one hour and 20 minutes.
