Lenglet, who spent last season on loan at Atleti, has made the move permanent ahead of the upcoming Club World Cup.
The 29 year-old signed a three-year deal with Diego Simeone's side, which he made 34 appearances for in the last campaign.
Atletico went on to finish third in LaLiga and were knocked out of the Copa del Rey semi-finals by Barcelona and the Champions League round of 16 by Real Madrid.
Lenglet spent the previous two seasons before that on loan in the Premier League with Tottenham and Aston Villa respectively.
The French defender played 160 times for Barcelona after his move from Sevilla in 2018, scoring seven goals in the process of winning the 2018-2019 LaLiga title, 2018-2019 Spanish Super Cup, and 2020-2021 Copa del Rey.
Lenglet made 16 appearances for France and was a part of their Nations League semi-final squad against Spain.
Atleti faces Paris Saint-Germain on 15 June in its first Club World Cup match, followed by group matches against Seattle Sounders and Botofogo.
