After Liam Oakes gave the hosts an early scare, Max Alleyne broke the deadlock in the 12th minute, pouncing on Jack Fitzwater's poor block to turn it home.
Rodri then doubled City's lead 12 minutes later with a stunning 25-yard effort, before a dismal five-minute period before the break for Exeter ultimately settled the game.
Fitzwater nodded a Rayan Cherki corner against an unfortunate Jack Doyle-Hayes and in, before putting an own goal against his name by deflecting Tijjani Reijnders's pullback past Joe Whitworth.
Semenyo, after his move from Bournemouth, came to the fore after the break, whipping in a lovely cross for Rico Lewis to tap home before coolly slotting City's sixth into the bottom-right corner 54 minutes in.
Reijnders bent a stunning strike from the edge of the box past Whitworth 71 minutes in, while Nico O'Reilly glanced a looping header over the goalkeeper 11 minutes from time.
Another debutant in Ryan McAidoo, scored City's ninth, with a venomous low drive from the edge of the box, before substitute George Birch got Exeter a stunning consolation by picking out the top-left corner from range in the 90th minute.
But Pep Guardiola's side had the final say as Lewis volleyed in from the right of the box in the first minute of stoppage-time to take City to double figures.
City's assistant coach Pep Lijnders, who was on the touchline with Pep Guardiola serving a one-match ban, is excited by what Semenyo can bring to City's attack.
"Antoine is settling in well," Lijnders said.
"He's a humble guy, we followed him for a long time, and he brings something to the frontline that we want and need.
"He can attack quickly, he wants to chase, he doesn't stop. You saw that he can adapt quickly to our style, so it's nice to have him with us.
"Overall, I think we have a really special group with experienced players that take the lead of the team – the way they guide the others.
"New players are settling in and the energy – I'm not just talking about the games but especially in training – it is special."
City now faces Newcastle United in the first leg of their Carabao Cup semi-final tie on Wednesday (AEDT), with Lijnders acknowledging the domestic cups are a priority for this season.
"We want to reach finals this year, and we're on our way, playing the semi-final in the League Cup. We want to go further in the FA Cup, and we are in the Champions League, and in a good position," he said.
"We want to build on that. The most important thing, Pep says it all the time... is that we want to grow and make the next step as a team.
"We need to keep working hard and take nothing for granted. We'll approach the next game as a final, and that will be Newcastle. With tha,t we will grow and make the next step."
